Bacteriophages (phages) are viruses that infect bacteria, distinguished by their host cell specificity, abundance, and diversity. They are increasingly studied as alternatives to antibiotics in combating antimicrobial-resistant bacteria and play significant roles in horizontal gene transfer and bacterial evolution. Despite their increasing recognition across various fields, their acquisition method has remained largely unchanged from the plaque assay method over the years. Here, I introduce two new methods for bacteriophage screening using water-in-oil droplets. Both approaches involve co-cultivation of bacteriophages and host cells within droplet microenvironments. One method detects fluorescently labeled phages, while the other labels host cells with fluorescence. Both methods enable the isolation of lytic phages; however, their suitability depends on the specific objectives of phage-related research. For example, the host cell staining method facilitates the detection and isolation of phages that do not form clear plaques, whereas fluorescent labeling of phages enables phage detection in environments lacking living host cells, such as cell-free phage assembly systems.
These complementary methods broaden the toolkit for phage screening and possibly engineering, enabling researchers to select the most effective approach for their specific project goals and experimental contexts.
